Description:
Fe65 (Amyloid beta A4 precursor protein-binding family B member 1) is an adaptor protein that forms a transcriptionally active complex with the gamma-secretase-derived amyloid precurson protein (APP) intracellular domain. APP functions as a cystosolic anchoring site that prevents Fe65 nuclear translocation. Following exposure to DNA damaging agents, Fe65 is released from the membrane and translocates to the nucleus to induce apoptosis. It acts by binding to Tyr-142-phosphorylated histone H2AX at double-strand breaks (DSBs), recruiting other pro-apoptosis factors such as MAPK8/JNK1. Fe65 is required for histone H4 acetylation at DSBs. Its ability to specifically bind modified histones and chromatin modifying enzymes such as KAT5/TIP60 probably explains the transcription activation activity of Fe65. Fe65 plays a role in the pathogenesis of Alzheimer's disease. It also blocks cell cycle progression by down-regulating thymidylate synthase expression.
