Description: Recombinant human PDGF-BB protein with molecular weight 28 kDa.
Background:
Platelet-derived growth factor, beta polypeptide (B Chain), PDGFB is a protein that in humans is encoded by the PDGFB gene. PDGF may be involved in the pathogenesis of a variety of disorders including atherosclerosis and certain types of cancer. Two PDGF-B chain residues, arginine 27 and isoleucine 30, have been identified by a site-directed mutagenesis programme. Substitutions in these positions can lead to PDGF mutants defective in both receptor affinity and cell activation as judged by displacement of PDGF-BB, mitogenic assay and inositol lipid turnover. PDGF-BB and IL-1B cooperate in inducing a sustained phosphorylation of PDGF receptor (PDGFR)-beta and its association with IL-1 receptor (IL-1R1) and inhibiting the polymerized collagen-induced serum response factor DNA binding activity in the nucleus, and this effect was mediated by the PDGFR-beta/IL-1R1 association and phosphatidylinositol 3-kinase/Akt/p70S6K pathway.
