Ampicillin is a β-lactam antibiotic with an amino group side chain attached to the penicillin structure, 6-amino-penicillanic acid. A semi-synthetic penicillin derivative that inhibits bacterial cell-wall synthesis (peptidoglycan cross-linking) by inactivating transpeptidases on the inner surface of the bacterial cell membrane. Ampicillin is more active against Gram-negative bacteria and is bactericidal only to growing Escherichia coli. Resistance is mediated by cleavage of β-lactam ring of ampicillin by β-lactamase. Recommended for antibacterial use in cell culture media and selection of resistant bacteria. Used as a selective agent for plasmid transformation such as pBR322 and pUC derivatives.
BSA (bovine serum albumin) is conjugated to ampicillin (1:179). The carboxyl group in ampicillin is directly linked to an amine group in BSA, and/or carboxyl group in BSA is directly linked to the amine group in ampicillin. Ampicillin has a molecular weight of 371.4 Da and BSA has a molecular weight of 66.4 kDa. The resultant conjugation solution is then buffer-exchanged with 20 mM PBS at a pH of 7.4.
Conjugated BSA-ampicillin is used to recognize ampicillin-specific antibodies by ELISA and lateral flow based immunoassay. Product can be stored below -20°C for up to 1 year.
