Protein Family: Cytokines
Pathway and Disease: Signal Transduction, Signaling Molecules and interaction, Neurodegenerative Disorders, Nervous and Sensory Systems
Alternate Names: Brain-derived neurotrophic factor, BDNF, AbrineurinDescription:
BDNF is a member of the nerve growth factor family of trophic factors. In the brain BDNF has a trophic action on retinal, cholinergic, and dopaminergic neurons, and in the peripheral nervous system it acts on both motor and sensory neurons. Some protein domains of BDNF are identical with those of NGF and another neurotrophic factor, designated NT-3 (neurotrophin-3 ). Polyclonal antibodies raised against murine NGF have been shown to cross-react with both NT-3 and BDNF. Recombinant Human Brain-Derived Neurotrophic Factor produced in E. coli is a non-glycosylated homodimer containing 2 x 119 amino acids with a MW of 26,984 Da.
Source: E. coli. Endotoxin level as measured by LAL is <0.01ng/ug or <0.1EU/ug.
Applications: E, WB
Application Notes:
BDNF is fully biologically active when compared to standard. The ED50 is calculated by the dose-dependent induction of ACHE (acetylcholine esterase) in rat basal forebrain primary septal culture with a range of 20 - 40 ng/ml.
Accession No.: P23560
MW: 26984 Da
Sequence:
MHSDPARRGE LSVCDSISEW VTAADKKTAV DMSGGTVTVL EKVPVSKGQL KQYFYETKCN PMGYTKEGCR GIDKRHWNSQ CRTTQSYVRA LTMDSKKRIG WRFIRIDTSC VCTLTIKRGR
Purity:
Purity > 97% as determined by RP-HPLC and reducing and non-reducing SDS-PAGE. Protein Content determined by UV spectroscopy at 280 nm and analysis by RP-HPLC calibrated against a known standard. Quantitation on SDS-PAGE against a known standard.
Format:
Each vial contains 10 ug of lyophilized protein. Reconstitute with 0.1 ml sterile deionized water for a final concentration of 0.1 mg/ml.
Storage: Store at -20°C. Product is guaranteed one year from the date of shipment. Following reconstitution, store at -20°C. We recommend to add a carrier protein (0.1% HSA or BSA) for long term storage.
