Background Information:
CD16a (FCGR3A) and its homolog CD16b are receptors for the Fc portion of human IgG. In contrast to CD64, which is a high affinity Fc binding protein, both CD16a and CD16b can bind Fc portion of human IgG with relative low affinity. Human CD16a is expressed mainly on natural killer cells, macrophages, T cells, and monocytes. While human CD16b is expressed on Neutrophils and eosinophils. In human cells, a single nucleotide polymorphism (T230A) creates a high binding (176V) and a low binding (176F) variant. Clinically, mutations in CD16a have been linked to vulnerability to viral infections, alloimmune neonatal neutropenia, and systemic lupus erythematosus.

Human CD16a protein (accession number NP_P08637-1, natural variant Val176) (Gly17-Gln208) with C-terminus His tag and Avi tag is expressed in HEK293 cells. This protein was site specifically labeled with Biotin by BirA ligase.
Molecular Weight:
The 227 amino acid protein has the predicted molecular weight of 25.5 kDa. The protein migrates at approximately 40-50 kDa on SDS-PAGE with DTT-reduced condition.
Purity: >95% based on SDS-PAGE
Formulation: 1xPBS buffer, 0.22 µm filtered
Protein Concentration:
25µg size is bottled at 0.2mg/mL concentration. 100 µg size is bottled at lot specific concentration.
Storage and Handling:
Quick spin the vial after receiving. Unopened vial can be stored at 4°C for 2 weeks or at -20°C and below for six months. The protein can be further diluted to 0.1 mg/mL with 0.22 µm filtered Tris PH 7.5 buffer. This stock solution should be aliquoted and stored at ≤ -70°C to minimize the freeze/thaw cycles. Carrier protein such as FBS and BSA need to be added if further dilution is needed.
