Glial cell line-derived neurotrophic factor (GDNF) is a neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake. Defects in GDNF are a cause of Hirschsprung disease, a lack of ganglions in the hindgut. GDNF is a disulfide-bonded homodimeric glycosylated protein of 134 amino acids. Recombinant Human GDNF produced in E. coli is a non-glycosylated homodimer containing 2 x 135 amino acids with a MW of 30,360 Da.

Application Notes:
Glial-Derived Neurotrophic Factor is fully biologically active when compared to standard. The ED50, calculated by the dose-dependent dopamin uptake in rat mesencephalic cultures was found to be 4-8 ng/ml.

Purity > 98% as determined by RP-HPLC and reducing and non-reducing SDS-PAGE. Protein Content determined by UV spectroscopy at 280 nm and analysis by RP-HPLC calibrated against a known standard. Quantitation on SDS-PAGE against a known standard.
Format:
Each vial contains 10 ug protein lyophilized in 10 mM sodium citrate, 100 mM NaCl, pH 4.0. Reconstitute with 0.1 ml sterile deionized water for a final concentration of 0.1 mg/ml.
Storage: Store at -20°C. Product is guaranteed one year from the date of shipment. Following reconstitution, store at -20°C. We recommend to add a carrier protein (0.1% HSA or BSA) for long term storage.
