Haptoglobin combines with free plasma hemoglobin, preventing loss of iron through the kidneys and protecting the kidneys from damage by hemoglobin, while making the hemoglobin accessible to degradative enzymes. Haptoglobin is a tetramer of two alpha and two beta chains. Recombinant haptoglobin beta-chain comprises a 245 amino acid fragment (162-406) corresponding to the mature haptoglobin beta-chain and is expressed in E. coli with an amino-terminal hexahistidine tag.
