MYB codes for a transcription factor protein that plays a major role in hematopoiesis and cell renewal in the colon lining. The gene is highly active in newly formed epithelial, endothelial and hematopoietic cells, then plays a lesser role as cells mature1.

Although MYB overexpression has been identified in pancreatic, colon and breast tumors, its translocations have only recently been indicated as having oncogenic effects2. Perhaps the most clinically relevant rearrangement discovered so far is the t(6,9) translocation, in which MYB is transferred to the NFIB gene on chromosome 91. This rearrangement occurs in about a third of adenoid cyst carcinoma tumors, and makes use of the DNA binding and transactivation sites of MYB (maintained in the new fusion gene) to act on MYB effector genes1.

Because MYB has such an extensive, varied list of target genes – including those that regulate core processes like cell development, cell adhesion, and angiogenesis – aberrations in its normal gene directing can lead to severe breakdowns in cell cycle regulation1. Additionally, MYB’s 3’ UTR is lost during translocation, which may serve to further compromise gene function, as the 3’ UTR is a binding site for MYB down-regulating miRNA’s3

MYB will undoubtedly continue to be an important biomarker for adenoid cyst carcinoma, helping to distinguish ACC from its histologically and morphologically similar mimics. Uncovering the specific oncogenic mechanisms of MYB translocation will further aid in diagnosis of ACC and other MYB-related cancers.
